In fact, the role of Ku70/80 proteins is highly complex and integrates different aspects of DNA repair. It has been shown that the Ku70/80 protein functions as a heterodimer to repair DSBs, with the two constituents stabilizing each other (Smider et al., 1994). Accordingly, these results may be attributed to a decrease in protein levels of both OsKu80 and OsKu70 caused by the down‐regulation of the expression of OsKu80 mRNA in KD‐Ku80 rice calli 2021-03-07 · Ku70/80 interacts directly with the RNA component of human telomerase, independent of the human telomerase reverse transcriptase protein. Ku antigen interacts with RBP-Jkappa and NF-kappaB p50 may act as a positive regulator of p50 expression in gastric cancer AGS cells. Ku70-positive cells were present in 90.5% of tumours without and in 80% of tumours with lymphatic metastases. A similar relationship was found for Ku80 expression. 2019-05-17 · The Ku70/80 heterodimer protein possesses a ring-shaped structure with high affinity for DSBs.

Ku70, a known nonhomologous end-joining (NHEJ) factor, also functions in tumor suppression, although this molecular mechanism remains uncharacterized. The protein forms a ring that can thread onto a DNA end, which explains its inability to bind to circular DNA molecules or internal positions on the chromosome [5].

Work in our lab has shown that a D192A/D195R mutation in helix five of the Ku70 von Willebrand A (vWA) domain leads to an NHEJ defect. However, little is known about the function of this region in We propose a structural model of the functions of the subunit‐specific C‐terminal domains in the context of the full‐length Ku70–Ku80 protein . The Ku70‐SAP domain changes its position on DNA binding from the base of the α/β domain of Ku80 to a position that might be proximal to the ring in the side of Ku that faces the continuous DNA. The heterodimer Ku70/80 plays a critical role in DNA repair and cell death induction after damage.

Ku70/Ku80 bindet an freie DNA-Enden und ist in die Reparatur von Doppelstrangbrüchen durch die nicht-homologe Endverknüpfung involviert. 2013-05-01 · When both Ku70/80 and RECQ1 were incubated with fork duplex in the presence of ATPγS, we observed a super-shifted species that is distinct from those obtained with binding of the fork DNA with Ku70/80 or RECQ1 alone (Fig. 3A, lane 3), indicating that Ku70/80 and RECQ1 can together bind the fork DNA structure.

et al. (2012) explored the role of Ku70/80 more closely and found that the C-terminal region of the Ku80 subunit is the main structural element that forms the interactions important to the function of Ku70/80 in bringing together the broken DNA ends. The data from Bennett et al. is consistent with the crystal structure as determined by Walker

As DNA end joining would be detrimental to chromosome ends, it was though that Ku has to be kept away from telomeres to prevent deleterious telomere-telomere fusion. This study highlights Ku70 as an important player not only in maintaining genomic stability through NHEJ-dependent functions, but also in regulating pancreatic β-cell proliferation, a novel NHEJ-independent function. Indeed, Ku70/80 has been proposed to perform at least two functions at telomeres.

The connection between the Ku70 level and stabilization of β-catenin merits further investigation, which could shed light on possible ways to enhance pancreatic β-cell proliferation. The heterodimer Ku70/80 plays a critical role in DNA repair and cell death induction after damage. The aberrant expression and localization of these proteins fail to control DNA repair and apoptosis. sClusterin is the Ku70 partner that sterically inhibits Bax-dependent cell death after damage in some pathologic conditions. Here we report that KU70 and DNA-PKcs unexpectedly function together during the induction of apoptosis after exposure to high levels of etoposide. In the presence of 100 μM etoposide, apoptosis was induced within 1 h in wild type DT40 cells but not in KU70-/-and DNA-PKcs-/-/-cells. In addition, the DNA-PK inhibitors NU7026 and wortmannin, as well as the caspase inhibitor Z-VAD-FMK, inhibited etoposide-induced apoptosis in wild type cells.

The data from Bennett et al. is consistent with the crystal structure as determined by Walker et al. (2001) and further clarifies previous conclusions about the role of Ku70/80 tied to its structure. 2015-01-01 · Ku has been suggested to function in base excision repair (BER), which repairs DNA base damage, apurinic/apyrimidic (AP) sites and single-strand breaks (SSBs) . Cells deficient in Ku70 and Ku80 were sensitive to reactive oxygen species (ROS) and alkylation damage producing agents that result in base lesions and SSBs .
